Introduction & Hypothesis

 

Immune checkpoint inhibitors (ICI) have transformed the treatment landscape for metastatic renal cell carcinoma (mRCC), yet responses remain heterogeneous, and no reliable biomarkers are currently used in clinical practice. While PD-L1 is a well-known marker, the research team hypothesized that lymphocyte activation gene-3 (LAG-3)—an inhibitory receptor that suppresses T-cell activation—might serve as a critical indicator of the tumor immune microenvironment (TME). 

 

Using Tempus Lens to analyze a large cohort of mRCC patients treated with frontline immunotherapy, the study aimed to characterize LAG-3 expression patterns, correlate them with genomic and immune features, and determine if LAG-3 could predict therapeutic response or survival.
